Description

The NucleoMag RNA Blood Kit is intended for the purification of RNA from whole blood samples collected in EDTA / Citrate blood collection tubes or in SARSTEDT® S-Monovette RNA Exact, Zymo DNA/RNA Shield™ Blood Tubes, or Tempus™ RNA Blood Tubes. With a focus on simplicity and efficiency, this kit eliminates the need for reagent removal and pelleting*, ensuring smooth processing. The purified RNA obtained from this kit is suitable for a range of downstream applications including RT-PCR and sequencing. The kit can be used either manually or automated on standard liquid handling instruments or automated magnetic separators. Included in the kit are essential components such as NucleoMag B-Beads, Buffers, Liquid Proteinase K, and RNase-free rDNase, ensuring reliable and high-quality RNA extraction. Note: Blood collection and stabilization tubes are not provided and must be purchased separately. *Processing of EDTA / Citrate or Tempus RNA Blood Tubes necessitates additional equipment and buffers as detailed in the user manual for comprehensive guidance.

Blood specimens sourced from two distinct donors were collected using SARSTEDT S-Monovette® RNA Exact or Zymo DNA/RNA Shield™ Blood Tubes, followed by purification employing the NucleoMag RNA Blood Kit via automated protocols on the MagnetaPure 32 Plus or KingFisher® Duo platform (n=3 per tube and instrument). Purified RNA underwent UV spectrometry analysis to assess total yield (blue bars) and purity (A260/280, orange dots). The data underscore remarkable reproducibility in RNA yield and purity across different donors, blood collection tubes, and automation platforms.



Robust RNA integrity profiles in S-Monovette® RNA Exact-collected samples

Blood specimens were procured from two distinct donors utilizing SARSTEDT S-Monovette® RNA Exact tubes and subjected to purification employing the NucleoMag RNA Blood kit via the automated MagnetaPure 32 Plus magnetic-rod extractor. Processing occurred subsequent to storage at room temperature for 2 hours (T0), 5 days (D5), or following two weeks of refrigeration at 2–8°C. Analysis conducted utilizing the Agilent Pico-Chip revealed excellent RNA integrity (8.0 ± 0.19) persisting across all examined time points and donors (n=3 for each donor and time point).

Blood specimens obtained from three diverse donors were collected utilizing the SARSTEDT S-Monovette® RNA Exact Blood Tube, followed by purification employing the NucleoMag RNA Blood Kit through automated protocols on either the MagnetaPure 32 Plus (MP 32 Plus) or the KingFisher® Duo (KF Duo) platform. Subsequent to purification, RNA samples underwent qRT-PCR analysis employing a Taqman® Probe for a 73 bp Actin amplicon (blue squares) and a 357 bp FOS amplicon utilizing the SensiFast™ Probe Lo-ROX One-Step Kit from Bioline on an Applied Biosystems® 7500 Real-Time PCR System. The data reveal consistent RNA performance across multiple donors and automation platforms.



Comprehensive Evaluation of RNA Integrity and Performance from Tempus™ RNA Blood Tubes

Blood specimens were obtained from six distinct donors using Tempus™ RNA Blood Tubes and subjected to purification utilizing the NucleoMag RNA Blood kit via automated protocols on the KingFisher® DUO (left) or the MagnetaPure 32 Plus (right) magnetic-rod extractors. Subsequent analysis included UV spectrometry (A) for total yield (blue bars) and purity (A260/280, orange dots). Additionally, RNA integrity was assessed using UV spectrophotometry with the Agilent Pico-Chip, revealing exceptional integrity (7.7±0.5) across all samples and donors. Furthermore, purified RNA underwent qRT-PCR analysis (B) employing Taqman® Probes for 73 bp Actin and 357 bp FOS amplicons with the SensiFast™ Probe Lo-ROX One-Step Kit on an Applied Biosystems® 7500 Real-Time PCR System. The findings demonstrate consistent RNA quality and performance across diverse donors and automation platforms.

*The Tempus™ RNA Blood  tubes require additional up-front handling and RNA pelleting at 4°C. Additional buffers must be orderd separately
