The UltraMarathonRT® Two-Step RT-PCR Kit is a next-generation solution for researchers needing sensitive and faithful RNA-to-cDNA conversion followed by amplification in RT-PCR experiments, regardless of RNA template length, structure, or GC content.
The kit is designed for experiments requiring high sensitivity and precision, significantly enhancing gene detection with lengths ranging from hundreds of nucleotides to tens of thousands of nucleotides. Unlike other RT-PCR kits, the UltraMarathonRT RT-PCR Kit requires only 0.1 pg of RNA input for the amplification of RNAs > 10 kb.
UltraMarathonRT is an ultra-processive reverse transcriptase enzyme with intrinsic helicase activity that actively unwinds RNA secondary and tertiary structures naturally without heat, faithfully producing full-length cDNAs from complex, long, or structured RNA templates that standard reverse transcriptases often miss. The DNA polymerase provided in this kit has high fidelity and is extremely processive in amplifying long cDNA (up to 30 kb) generated by UltraMarathonRT.

Reagents required but not supplied: RNA template: 0.1 pg-2 μg total cellular RNA or 0.1 pg-500 ng of mRNA are recommended for each reaction. If target template is > 10 kb, a minimum input of 1 pg is recommended for both total cellular RNA or mRNA.
Storage conditions: For short duration of storage (< 3 month), UltraMarathonRT can be placed in a -20℃ freezer. -80℃ is recommended for long-term storage. Note that UltraMarathonRT can endure 10 freeze-thaw cycles without losing noticeable activity. After 20 freeze-thaw cycles, UltraMarathonRT maintains 90% of its original activity.

Amplification of transcripts from 1.2 kb to 14.9 kb using 100 ng Hela total RNA. The cDNA’s generated by Induro RT were amplified by LongAmp Taq. Reverse transcription was performed using Oligo(dT) 18 primer.
