Description
The UltraMarathonRT® (uMRT) cDNA Synthesis & Amplification Kit enables you to see more of the transcriptome in each sample. uMRT is a next-generation reverse transcriptase that synthesizes end-to-end cDNA in a single pass from even the most complex RNA molecules—long, structured, high-GC, or low-abundance.
Conventional retroviral RTs are ‘distributive’ meaning they attach and detach many times from each RNA transcript which often creates truncated, incomplete, or incorrect cDNAs. This is often caused by secondary structures or repeat sequences within the template.
UltraMarathonRT solves this problem. It is a highly engineered group II intron-encoded reverse transcriptase with intrinsic helicase activity. It bypasses the drop-offs, biases, and blind spots common in traditional cDNA synthesis workflows.

Storage Conditions: For short duration of storage (< 6 month), UltraMarathonRT can be stored in a -20℃ freezer. -80℃ is recommended for long-term storage. All other kit components are stable in a -20℃ freezer for long-term storage. Note that UltraMarathonRT can endure 10 freeze-thaw cycles without losing noticeable activity. After 20 freeze-thaw cycles, UltraMarathonRT maintains 90% of its original activity.

RNA-seq is a powerful method for studying gene expression, but commonly used retroviral reverse transcriptases (e.g., MMLV and AMV) lack two critical properties for effective reverse transcription: intrinsic helicase activity, which enables the unwinding of complex RNA secondary structures, and high processivity, which allows for the synthesis of full-length cDNA from long RNA molecules.
UltraMarathonRT (uMRT) is a reverse transcriptase enzyme designed to overcome these limitations and improve RNA coverage. The cDNA Synthesis & Amplification Kit, powered by uMRT, combines intrinsic helicase activity with ultraprocessivity to efficiently reverse transcribe long RNA sequences (>30 kb—compared to the ~2 kb typically transcribed by retroviral RTs), as well as RNA with high GC content, complex structures, or low abundance. uMRT also functions at lower temperatures, helping preserve RNA integrity and enabling a more comprehensive view of the transcriptome.
