Description
The DepleteX® Globin Depletion Kit selectively removes highly expressed globin transcripts (HBA1, HBA2, HBB, & HBD) in vitro before sequencing. This process redirects sequencing resources toward unique biologically relevant transcripts, maximizing UMI and gene sensitivity.
For research use only. Not for use in diagnostic procedures.
Overview
Enhance RNA-Seq quality by eliminating globin fragments.
Apart from ribodepletion, RNA samples often contain a limited number of well-characterized, highly expressed transcripts. However, resequencing these transcripts contributes minimally to biologically relevant information. The dominance of highly expressed transcripts hinders coverage for low-expressing counterparts, resulting in reduced sensitivity for detecting disease-related low-expressing genes, rare variants, and unique fusion events.
Globin mRNA dominates the mRNA content of red blood cells and whole blood. Three globlin genes (HBA1, HBA2 and HBB) constitute ≥30% of total mRNA from blood.
Key highlights:
- Unbiased removal of human globin from RNA-seq libraries
- Achieve >99% depletion of contaminating globin
- Rapid one-hour depletion incubation
